FlexiPro TFF enables rapid and complete diafiltration while maintaining particle integrity

This application note showcases the low-shear capabilities of our single-use FlexiPro TFF system. This collaboration with Meissner Filtration using mRNA-loaded LNPs from Inside Therapeutics demonstrated rapid and complete diafiltration while maintaining particle integrity.

Short summary:

• No measurable degradation of mRNA-loaded LNPs
• Particle integrity was evaluated using poly dispersion index (PDI) and mean volume diameter
• Complete buffer exchange in 3.2 DVs (diafiltration volumes) compared to average 5-10 DV for mRNA-loaded LNPs
• Moreover, LNP integrity was maintained even after challenging the FlexiPro TFF system by doubling the processing volume to 6 DV
• The study used the Very Low Flow Kit (VLFK) with 1/8” tubing and a flow rate of 1-30 L/hr
